He was the son of a musician and grew up in a musical family. His father, Giuseppe Cannio, was a composer and conductor, and his mother, Maria Luisa, was a singer. Enrico showed an early interest in music and began studying piano and composition at a young age. Cannio's musical education began at the Conservatory of San Pietro a Majella in Naples, where he studied under the guidance of some of the most prominent Italian composers of the time, including Paolo Serrao and Beniamino Cesi. He graduated from the conservatory in 1895 with a degree in piano and composition. After completing his studies, Cannio began working as a composer and conductor in Naples. He quickly gained a reputation as a talented composer and was soon commissioned to write music for various theatrical productions. In 1901, he composed his first operetta, "La Maschera di Ferro," which was a great success and established him as a leading composer of light opera. Over the next few years, Cannio continued to compose operettas and other works for the stage. In 1904, he wrote "La Bella Napoli," which became one of his most popular works and was performed all over Italy. The operetta was set in Naples and featured a number of traditional Neapolitan songs, which helped to popularize the music of the region. In 1906, Cannio moved to Milan, where he continued to compose for the stage. He wrote a number of successful operettas, including "La Signorina Papillon" and "La Vedova Allegra," which were performed in theaters throughout Italy and Europe. In 1910, he composed "La Canzone del Sole," which became one of his most famous songs and was later recorded by a number of popular singers. During World War I, Cannio served in the Italian army as a musician. He wrote a number of patriotic songs and marches, including "La Leggenda del Piave," which became a popular anthem for the Italian soldiers. After the war, he returned to composing for the stage and wrote a number of successful operettas, including "La Principessa della Czarda" and "La Danza dei Milioni." In addition to his work as a composer, Cannio was also a respected conductor. He conducted many of his own works and also worked with other composers, including Giacomo Puccini and Pietro Mascagni.
