He received his early education in nearby Korbach and then went on to study theology at the University of Erfurt. After completing his studies, Nicolai became a Lutheran pastor and served in several churches in Germany. He was known for his eloquent preaching and his devotion to the Lutheran faith. In 1586, he was appointed as the pastor of the small town of Unna, where he would spend the rest of his life. Nicolai was also a talented musician and composer. He wrote several hymns and chorales that became popular in Lutheran churches throughout Germany. His most famous work is the hymn "Wachet auf, ruft uns die Stimme" ("Wake, Awake, for Night is Flying"), which is based on the parable of the ten virgins from the Gospel of Matthew. The hymn is still sung in churches today and is considered one of the greatest works of Lutheran hymnody. In addition to his hymns, Nicolai also composed several works for organ and other instruments. His music is characterized by its simplicity and elegance, and it reflects his deep faith and devotion to God. One of Nicolai's most famous works for organ is the chorale prelude "Wie schön leuchtet der Morgenstern" ("How Brightly Shines the Morning Star"). The piece is based on a hymn by the same name and is considered one of the greatest works of the Baroque period.
