He was born in Mölln, a small town in the Duchy of Lauenburg, which is now part of Germany. Müthel was the son of a Lutheran pastor, and he showed an early interest in music. Müthel's musical talent was recognized at an early age, and he was sent to study with Johann Sebastian Bach in Leipzig when he was just 14 years old. Bach was impressed with Müthel's abilities and took him on as a student. Müthel studied with Bach for two years, during which time he learned a great deal about composition and keyboard playing. After leaving Bach's tutelage, Müthel went on to study with other prominent musicians of the time, including Johann Joachim Quantz and Johann Adolph Hasse. He also spent time in Copenhagen, where he studied with Johann Adolph Scheibe. Müthel's travels and studies allowed him to develop a unique style that blended elements of the Baroque and Classical periods.
