His father was a clarinetist, and his mother was a pianist, which influenced his interest in music from a young age. Deffayet began his musical journey by learning the clarinet at the age of six. However, he later switched to the saxophone, which became his primary instrument. He studied at the Paris Conservatory, where he was a student of Marcel Mule, a renowned saxophonist and teacher. Deffayet was an exceptional student and won several awards during his time at the conservatory. After completing his studies, Deffayet began his professional career as a saxophonist. He joined the French National Radio Orchestra in 1947 and became its principal saxophonist in 1950. He also performed with other orchestras, including the Paris Opera Orchestra and the Lamoureux Orchestra. Deffayet was known for his exceptional technique and musicality, which made him one of the most sought-after saxophonists of his time. In addition to his career as a performer, Deffayet was also a conductor. He founded the Ensemble Instrumental de France in 1969, which was a chamber orchestra that specialized in contemporary music. He also conducted several other orchestras, including the Orchestre National de France and the Orchestre de Paris. Deffayet was also a dedicated teacher and taught at the Paris Conservatory from 1956 to 1986.
