He was born in Turin, Italy, to a family of musicians. His father, Carlo Sinigaglia, was a composer and conductor, and his mother, Maria Sinigaglia, was a pianist. Leone showed an early interest in music and began studying the violin at the age of six. He later studied composition with Giovanni Bolzoni and Luigi Torchi. Sinigaglia's early compositions were influenced by the German Romantic tradition, particularly the music of Brahms and Wagner. He also drew inspiration from Italian folk music and the works of Italian composers such as Verdi and Puccini. In 1890, he won the prestigious Sonzogno Prize for his opera "Il Vassallo di Szigeth," which was based on a historical event in Hungary. Sinigaglia's career as a conductor began in 1892, when he was appointed the conductor of the orchestra at the Teatro Regio in Turin. He also conducted the orchestra at La Scala in Milan and the Philharmonic Orchestra of Berlin. In addition to his work as a conductor, Sinigaglia was a prolific composer, writing works for orchestra, chamber ensembles, and solo instruments. One of Sinigaglia's most famous works is his Symphony No. 2 in F major, which was premiered in 1908. The symphony is notable for its lush harmonies and sweeping melodies, which reflect Sinigaglia's Romantic sensibilities.
