His music was characterized by its expressive melodies, intricate harmonies, and use of chromaticism. Milanuzzi was particularly skilled at setting texts to music, and his works were praised for their clarity and emotional depth. One of Milanuzzi's most famous works is his opera La Vita Humana, which was first performed in Venice in 1630. The opera tells the story of a young man who is torn between his love for a woman and his duty to his family. The music is rich and complex, with arias, duets, and choruses that showcase Milanuzzi's skill as a composer. Milanuzzi's other notable works include his collection of madrigals, Il Primo Libro de Madrigali a Cinque Voci, which was published in 1628. In addition to his compositions, Milanuzzi was also known for his work as a music theorist.
